چکیده

OBJECTIVE Hemoglobin (Hb) S disorder [beta6(A3)Glu-->Val, GAG-->GTG] is an important hemoglobinopathy with the highest endemicity in Africa. METHODS Here, the author performs a basic bioinformatics gene ontology study to assess the effect of co-expression between nucleic acid sequence for human Hb S beta globin chain and U7.623. RESULTS According to this study, the analytical results show that fully recovery of hemoglobin in both function and biological process can be got. CONCLUSION Here, the author proposed that U7 snRNA might be a probable tool for treatment by gene therapy approach in Hb S disorder.

Cloning and characterization of the Drosophila U7 small nuclear RNA.

Base pairing between the 5' end of U7 small nuclear RNA (snRNA) and the histone downstream element (HDE) in replication-dependent histone pre-mRNAs is the key event in 3'-end processing that leads to generation of mature histone mRNAs. We have cloned the Drosophila U7 snRNA and demonstrated that it is required for histone pre-mRNA 3'-end processing in a Drosophila nuclear extract. The 71-nt Dro...

FUS/TLS contributes to replication-dependent histone gene expression by interaction with U7 snRNPs and histone-specific transcription factors.

Replication-dependent histone genes are up-regulated during the G1/S phase transition to meet the requirement for histones to package the newly synthesized DNA. In mammalian cells, this increment is achieved by enhanced transcription and 3' end processing. The non-polyadenylated histone mRNA 3' ends are generated by a unique mechanism involving the U7 small ribonucleoprotein (U7 snRNP). By usin...

The Cellular Processing Capacity Limits the Amounts of Chimeric U7 snRNA Available for Antisense Delivery

Many genetic diseases are induced by mutations disturbing the maturation of pre-mRNAs, often affecting splicing. Antisense oligoribonucleotides (AONs) have been used to modulate splicing thereby circumventing the deleterious effects of mutations. Stable delivery of antisense sequences is achieved by linking them to small nuclear RNA (snRNAs) delivered by viral vectors, as illustrated by studies...

Enhancement of β‐Globin Gene Expression in Thalassemic IVS2‐654 Induced Pluripotent Stem Cell‐Derived Erythroid Cells by Modified U7 snRNA

The therapeutic use of patient-specific induced pluripotent stem cells (iPSCs) is emerging as a potential treatment of β-thalassemia. Ideally, patient-specific iPSCs would be genetically corrected by various approaches to treat β-thalassemia including lentiviral gene transfer, lentivirus-delivered shRNA, and gene editing. These corrected iPSCs would be subsequently differentiated into hematopoi...

The U7 small nuclear RNA genes of Xenopus borealis.

Small nuclear RNA genes transcribed by RNA polymerase I1 have 3 conserved sequence elements necessary for efficient transcription. The distal sequence element, DSE (-250 to -200) is an enhancer containing at least one octamer sequence motif. The DSE has been shown to function independent of orientation and to some extent position [ I ] . The proximal sequence element, PSE (-60 to -50) is essent...
